Cytochrome c Interaction with Cardiolipin Plays a Key Role in Cell Apoptosis: Implications for Human Diseases

چکیده
In the cell cytochrome, c performs different functions depending on environment in which it acts; therefore, has been classified as a multifunction protein. When anchored to outer side of inner mitochondrial membrane, native cytochrome acts Schweitzer-StennerSchweitzer-Stenner that transfers electrons from reductase oxidase respiratory chain. On other hand, interact with cardiolipin (one phospholipids making up membrane) and form c/cardiolipin complex apoptotic process, protein reorganizes its structure into non-native state characterized by asymmetry. The formation is fundamental step pathway, since structural rearrangement induces peroxidase activity c, subsequent permeabilization release free cytoplasm, where activates process. Apoptosis closely related pathogenesis neoplastic, neurodegenerative cardiovascular diseases; this contest, biosynthesis remodeling are crucial for regulation Since role promoter apoptosis strictly depends conformation(s) acquires when bound such event leads traslocation cytosol, functional properties fate will be focus present review.

Innate Immunity Plays a Key Role in Leishmania Infection: Implications for Vaccine Design
Neutrophils are part of the first line of immune response and are essential for resistance against a variety of pathogens. They professionally mediate direct killing of pathogens, recruit other phagocytes by specific chemokines, produce cytokines and interact with different immune cells to shape the adaptive response. Leishmania as an obligatory intracellular parasite has evolved to benefit thi...
متن کاملCytochrome C interaction with cardiolipin/phosphatidylcholine model membranes: effect of cardiolipin protonation.
Resonance energy transfer between anthrylvinyl-labeled phosphatidylcholine as a donor and heme moiety of cytochrome c (cyt c) as an acceptor has been employed to explore the protein binding to model membranes, composed of phosphatidylcholine and cardiolipin (CL). The existence of two types of protein-lipid complexes has been hypothesized where either deprotonated or partially protonated CL mole...
متن کاملMrpL35, a mitospecific component of mitoribosomes, plays a key role in cytochrome c oxidase assembly
Mitoribosomes perform the synthesis of the core components of the oxidative phosphorylation (OXPHOS) system encoded by the mitochondrial genome. We provide evidence that MrpL35 (mL38), a mitospecific component of the yeast mitoribosomal central protuberance, assembles into a subcomplex with MrpL7 (uL5), Mrp7 (bL27), and MrpL36 (bL31) and mitospecific proteins MrpL17 (mL46) and MrpL28 (mL40). We...
متن کاملT cell-mediated Fas-induced keratinocyte apoptosis plays a key pathogenetic role in eczematous dermatitis.
Clinical and histologic similarities between various eczematous disorders point to a common efferent pathway. We demonstrate here that activated T cells infiltrating the skin in atopic dermatitis (AD) and allergic contact dermatitis (ACD) induce keratinocyte (KC) apoptosis. KCs normally express low levels of Fas receptor (FasR) that can be substantially enhanced by the presence of IFN-gamma. KC...
متن کاملConformational properties of cardiolipin-bound cytochrome c.
Interactions of cytochrome c (cyt c) with cardiolipin (CL) are important for both electron transfer and apoptotic functions of this protein. A sluggish peroxidase in its native state, when bound to CL, cyt c catalyzes CL peroxidation, which contributes to the protein apoptotic release. The heterogeneous CL-bound cyt c ensemble is difficult to characterize with traditional structural methods and...
